We have synthesized a 582,970-base pair Mycoplasma genitalium genome. This synthetic genome, named M. genitalium JCVI-1.0, contains all the genes of wild-type M. genitalium G37 except MG408, which was disrupted by an antibiotic marker to block pathogenicity and to allow for selection. To identify the genome as synthetic, we inserted "watermarks" at intergenic sites known to tolerate transposon insertions. Overlapping "cassettes" of 5 to 7 kilobases (kb), assembled from chemically synthesized oligonucleotides, were joined by in vitro recombination to produce intermediate (...) assemblies of approximately 24 kb, 72 kb ("1/8 genome"), and 144 kb ("1/4 genome"), which were all cloned as bacterial artificial chromosomes in Escherichia coli. Most of these intermediate clones were sequenced, and clones of all four 1/4 genomes with the correct sequence were identified. The complete synthetic genome was assembled by transformation-associated recombination cloning in the yeast Saccharomyces cerevisiae, then isolated and sequenced. A clone with the correct sequence was identified. The methods described here will be generally useful for constructing large DNA molecules from chemically synthesized pieces and also from combinations of natural and synthetic DNA segments. 10.1126/science.1151721. In this paper, we consider how dignity is discursively constructed in the context of work dominated by physicality and dirt. Based on semi-structured interviews with garbage workers, our analysis considers how the deprivations they experience are cast through discourses intended to construct their individual and collective worth. We consider the manner in which dignity maybe denied to such workers through popular repudiations of individuality and status. We demonstrate how this positioning arises from contact with physical dirt, and associations with socially (...) dirty work based on ascriptions of servility, abuse and ambivalence. We go on to consider how garbage workers respond to this positioning through discourses of ‘everyday heroism’. Heroism is evoked through three inter-related narratives that speaks to a particular type of masculinity. The first takes the form of a classic process of reframing and recalibration through which workers not only renegotiate their public position and status, but also point to the inherent value to be had in working with dirt as part of that which we identify as a process of ‘affirmation’. The second narrative arises from the imposition of favourable social and occupational comparisons that effectively elevate garbage collectors’ social position. The third discourse—and previously unobserved in respect of garbage work—centres on paternalistic practices of care. Combined, these discourses disrupt the generally held view that dirty work is antithetical to heroism and wounds dignity. (shrink)

In The Blue Book, Wittgenstein defined a category of uses of “I” which he termed “I”-as-subject, contrasting them with “I”-as-object uses. The hallmark of this category is immunity to error through misidentification (IEM). This article extends Wittgenstein’s characterisation to the case of memory-judgments, discusses the significance of IEM for self-consciousness—developing the idea that having a first-person thought involves thinking about oneself in a distinctive way in which one cannot think of anyone or anything else—and refutes a common objection to the (...) claim that memory-judgments exhibit IEM. This article serves as an introduction to the laws-of-biology debate. After introducing the main issues in an introductory section, arguments for and against laws of biology are canvassed in Section 2. In Section 3, the debate is placed in wider epistemological context by engaging a group of scholars who have shifted the focus away from the question of whether there are laws of biology and toward offering good accounts of explanation(s) in the biological sciences. Section 4 introduces two relatively new (...) pieces of science – metabolic scaling theory and ecological stoichiometry – that have not been topics of much discussion by philosophers but are relevant because they have at least some of the hallmarks of laws of nature. Section 5 concludes by pointing out that discovering laws of biology, if any there be, will not necessarily answer the questions raised by the debate in the first place: we will still want to know how biology compares to other sciences, how to characterize its systems and processes, and whether accounts in terms of laws always or usually constitute adequate explanations in various sciences. Theorists about love typically downplay the scale of persistent and possibly intractable disagreement about love. Where they have considered such disagreements at all, they have tended to treat them as an example of the lack of clarity surrounding the concept of love, a problem which can be resolved by philosophical analysis. In doing so, they invariably slip into prescriptive mode and offer moral injunctions in the guise of conceptual analyses.This article argues for philosophical modesty. I propose that the starting point (...) of any coherent philosophical investigation of love must be a willingness to take our disagreements seriously. These disagreements stem from profound moral differences: we disagree about love inasmuch as we disagree about how we should properly treat one another.With a series of examples drawn from philosophy, literature and real life I attempt to illustrate some of the disagreements that arise in relation to erotic love. Drawing upon the work of Wittgenstein, Friedrich Waissman and W.B. Gallie, I suggest that any robust theory of love needs to take account of its contestable nature and the integral role it plays in our moral life. (shrink)

Using an ethnographic and longitudinal interview study of college women and in-depth interviews with their parents, I argue that mid-tier flagship universities still push women toward gender complementarity—a gender-traditional model of economic security pairing a career oriented man with a financially dependent woman. Combining multilevel and intersectional theories, I show that the infrastructure and campus peer culture at Midwest University supports this gendered logic of class reproduction, which reflects an affluent, white, and heterosexual femininity. I argue that this logic may (...) only work for a minority of students, and plays a role in reinforcing class inequities among women. (shrink)

This essay reconstructs the medieval practice of evangelical poverty as a resource for contemporary political theology. Francis of Assisi and his predecessors committed themselves to a form of voluntary poverty that directly contested the distribution of social power in twelfth-century Europe. Evangelical poverty was for them a critical and liberating practice. Yet they disagreed about how this practice was related to standing norms of ecclesial authority. Francis broke with the earlier movements by defining evangelical poverty as a posture of humility (...) and obedience rather than as a counterclaim on apostolic authority. These movements are worth retrieving both for their shared commitment to a liberating poverty and for the questions they raise about the relationship between poverty and authority. (shrink)

The visual arts include painting, sculpture, photography, video, and film. But many people would argue that music is the universal or only art of sound. In the modernist era, Western art music has incorporated unpitched sounds or ‘noise’, and I pursue the question of whether this process allows space for a non-musical soundart. Are there non-musical arts of sound—is there an art phonography, for instance, to parallel art photography? At the same time, I attempt a characterization of music, contrasting acoustic, (...) aesthetic, and acousmatic accounts. My view is that there is some truth in all of these. I defend the claim that music is an art with a small ‘a’—a practice involving skill or craft whose ends are essentially aesthetic, that especially rewards aesthetic attention—whose material is sounds exhibiting tonal organization. But acoustic and acousmatic accounts help to distinguish between music and non-musical soundart, since music must have a preponderance of tones for its material. (shrink)

In this study, the author uses ethnographic and interview data from a women's floor in a university residence hall to examine how some heterosexual women's gender strategies contribute to their homophobia. The author describes a prevailing heterosexual erotic market on campus—the Greek party scene—and the status hierarchy linked to it. Within this hierarchy, heterosexual women assign lesbians low rank because of their assumed disinterest in the erotic market and perceived inability to acquire men's erotic attention. Active partiers invest more in (...) this social world and prefer higher levels of social distance from lesbians than do others. These women also engage in same-sex eroticism primarily designated for a male audience. They define their behaviors as heterosexual, reducing the spaces in which lesbians can be comfortable. Finally, the author concludes by discussing the unique nature of women's homophobia and the links between sexism and heterosexism. (shrink)

This paper develops two prescriptive standards — the Expectation and Reputation guidelines — that may help businesspeople avoid violating clearly accepted truth standards. The guidelines also assist in determining whether truth is required in circumstances where honesty seems in conflict with the practical demands of business. This article questions a number of widely held views of the role of values in psychotherapy. It begins with a discussion of the now largely discredited view that psychotherapy can be value free. It also broadens this challenge to question the popular idea that values form an inescapable part of the therapeutic encounter. While this view is correct in outline, it is necessary to reject the underlying conception of values as largely arbitrary preferences that the client and the therapist bring (...) to the encounter, as this fails to do justice to the inherently ethical nature of psychotherapy. It argues that we should recover the Greek notion of therapy as essentially concerned with the character of a person. In other words, the goal of therapy is virtue. (shrink)

This article introduces a novel theoretical framework for psychopathy that bridges dominant affective and cognitive models. According to the proposed impaired integration (II) framework of psychopathic dysfunction, topographical irregularities and abnormalities in neural connectivity in psychopathy hinder the complex process of information integration. Central to the II theory is the notion that psychopathic individuals are “‘wired up’ differently” (Hare, Williamson, & Harpur, 1988, p. 87). Specific theoretical assumptions include decreased functioning of the Salience and Default Mode Networks, normal functioning in (...) executive control networks, and less coordination and flexible switching between networks. Following a review of dominant models of psychopathy, we introduce our II theory as a parsimonious account of behavioral and brain irregularities in psychopathy. The II theory provides a unified theoretical framework for understanding psychopathic dysfunction and integrates principle tenets of affective and cognitive perspectives. Moreover, it accommodates evidence regarding connectivity abnormalities in psychopathy through its network theoretical perspective. (shrink)

In this article I explore Eve Garrard's recent account of evil and some work of Colin McGinn's on the same topic. I argue that neither provides a satisfactory account of evil. In doing so, I discuss the role of conscience, sadism and indifference to the suffering of others in evil-doing. I argue that the evil-doer can be admirable and I explore the relation between agent and action in the evil deed.The idea that evil is mysterious is considered and I conclude (...) with some comments on the relation between evil and the idea of a fellowship amongst human beings. If one sits in the stands for awhile at a local sporting contest, whether it is wrestling, soccer, baseball or particularly basketball, before long someone will exclaim toward a referee, ?That was a makeup call. You owe us one.? Everyone knows what this means but if an eight-year old beside you hears this screamed for the first time and asks, ?What does that mean?? An explanation given to her will be something like ?that's when an official makes a call and (...) immediately realizes it was a wrong call so at the first opportunity he can he makes a call against the team that has benefitted from the previous call; he is trying to even things out for his mistake.? The referee or umpire is attempting to be fair by ?giving back? the call with a ?makeup call.? A make-up call can be defined as the act of compensating for a questionable or bad officiating call by making a proportionally even call against the team that was aided by the first call. Usually these are immediate and obvious. They can also be conscious or subconscious, intentional or unintentional. The hope is that the two calls generally offset one another without either team being dramatically harmed. I will argue that the makeup call is prima facie immoral but if one were to attempt to find moral justification for it, this could best be done through a corrective theory of justice. But this presents a number of moral questions and ambiguities, most significantly, is a makeup call just and fair? Is it appropriate to understand the makeup call as an example of two wrongs making a right? A broader philosophic al question about officiating is whether each call in a contest should be viewed by as an independent single call with the goal of the official getting that one immediate call correct or should each call be understood as in a direct relationship with every other call in that game as a type of gestalt thus justifying makeup calls? This work will probe the depths of the highly suspect yet common moral puzzle, the makeup call. (shrink)

This article aims to contribute to the developing area of feminist scholarship on women and political violence, through a study of women in one of Europe's oldest illegal armed movements, the radical Basque nationalist organization ETA. By tracing the changing patterns of women's participation in ETA over the past four decades, the article highlights the historical factors that help explain the choice of a small number of Basque women to participate directly in political violence, and shows how these factors have (...) differed from those for men. While the gender politics of radical nationalism are intricately linked to cross-cultural associations of militarism with certain forms of masculinity, the article also stresses the importance of understanding women's activism in ETA in the context of the organization's characteristic as an ethnic nationalist movement, as well as the wider historical circumstances of the movement's development, including the modernization of Spanish and Basque society over the past four decades. Although comparisons with women in other armed movements are possible, such historical specificities undermine any attempt to construct a universal theory of women and ‘terrorism’, such as Robin Morgan's ‘couple terrorism’ thesis. Finally, the article examines the changing representations of female ETA activists in the Spanish and Basque media. Although women ETA activists are now regarded as ‘normal’, popular representations continue to link women's armed activism with deviant sexuality and the transgression of their natural destiny as mothers. The different treatment of women is evident as well in claims of sexual torture made by some detainees. The article concludes that although the participation of women in political violence poses disquieting questions for the largely anti-militarist women's movement, case studies of women in armed organizations, as well as their place in the wider practices of conflict, are an important contribution both to feminist debates about violence and to wider studies of political violence. This paper is an exploration and interpretation of Kierkegaard's account of Christian belief. I argue that Kierkegaard believed that the Christian metaphysical tradition was exhausted and hence that there could be no defence of belief in God in purely rational terms. I defend this interpretation against objections, going on to argue that Kierkegaard thought it possible to defend a post-metaphysical conception of religious belief. I argue that Kierkegaard thought that such a defence was available if we understand correctly what it (...) is to speak with ethico-religious authority. I argue that, when interpreted in the way I outline, Kierkegaard's notion of ethico-religious authority shows his conception of religious belief to have great plausibility. However, Kierkegaard goes on to argue that an individual's true relationship with God is constituted through the cultivation of guilt and the sense of himself as a sinner, and I give reasons for rejecting this claim, arguing that such cultivation is a form of asceticism. The language of structural sin is most often used to describe sin that inheres in laws, institutions, or social roles—in short, in the objective social architecture of our everyday lives. This article argues that structural sin should also be understood as including a subjective dimension, describing the determinate habits or dispositions instilled by sharing in the life of a particular society. Part of what is structured by structural sin, in other words, is agency itself. The reason that many theologians have (...) resisted this idea is that it seems to undermine the conditions of moral responsibility. If our capacities for knowing and loving the good are always already concretely misshapen, can we rightly be held accountable for what we do? This article argues further that we can, drawing on the work of Judith Butler to sketch a fresh account of personal responsibility in the face of structural sin. (shrink)

The McNaughton Rules, which are used when someone pleads insanity at the time of a homicide, are out of date and unsatisfactory. Suggestions have been made about how the insanity defence can be reformulated. The preference of a defence of diminished responsibility means abandoning an ancient and humane principle of not convicting those who are so mentally disordered as not to be responsible for their actions. There is a need for Parliament to consider changes to the law both to prevent (...) the mentally disordered being sent to prison inappropriately, and because the Mental Health Act 1983 has not taken account of rare cases where an offender such as an epileptic might be found legally insane but not mentally disordered. Integral Science provides the empirical rigor needed to shift medicine's worldview. The shift in science will give rise to Integral Medicine, which will emerge from the integration and transformational change of biomedicine, psychosocial approaches, Complementary and Alternative Medicine, and other reform movements. The root metaphor of Integral Medicine is a healthy and sustainable ecosystem. At its heart are mind-body holism and collaborative learning. The last several decades have seen the emergence of intellectual property rights (IPRs), especially patents, as a key issue in developments across the fields of law, the economy and the biosciences, and as part of the burgeoning "bioeconomy". This paper examines how the categories of nature and knowledge, so vital to IPR regimes that support bioeconomy-type projects, are challenged by the allegation of biopiracy. It reflects on the relationship between nature, IPR and the bioeconomy, and presents an example of how (...) the creation of new categories, in this case genetic resources, come to impact on how we come to understand nature in relation to the economy and IPRs. The paper then follows this with an analysis of how knowledge is treated in those same systems, focusing specifically on some of the challenges that arise when attempting to bring traditional knowledge (TK) under the rubric of the IPR system. Finally, it examines the allegation of biopiracy in greater depth, and reflects on how it critiques the way the bioeconomy and the IPR regime treat nature, culture, knowledge, and the relationship between them.
